Can a 2 door Jeep Wrangler work well for overlanding?

If it is just for 1 person maybe yes mate. You will struggle with storage space and payload. If you travel light you won't have issues though and will also be able to hit some really tough trails!
 
I would say yes but just for 1 person and some clever packaging. The only issue here is space; therefore, if you only take the essentials and package cleverly you will be fine
